DNA Sequencing


DNA hybridization relies on base-complement pairing

  • to produce the estimate of difference
  • short fragment of mismatch will not prevent hybridization
  • long mismatch or multiple mismatches will prevent hybridization
    Sequencing lists the nucleotides [bases] 5'-end to 3'-end
  • DNA from 2 sources can be compared to determine how different they are
    . . - defined as number of nucleotides different
    . . - or even, where they are different
  • with DNA sequences from any group of species
    . . - a genus, family, order...
    . . - nucleotides difference estimates evolutionary relationship
    . . . - if nucleotides change at constant rate:
    . . . . different (bases) = time since divergence
    . . . . overlap (bases) = percent relationship
